PROFESSIONAL EXPERTISE AND MAJOR ACHIVEMENTS
Nuclear transport in yeast S.cerevisiae, structure of the Nuclear Pore Complex (NPC), development of a novel methods to study NPC.
1. Defining minimal nuclear pore complex.
2. Novel method for in vivo measurements of the transport kinetics rates.
3. Novel method for in vivo measurements of the sieving potential of the NPC.
4. Discovery of the Ssa1p (Hsp70p of yeast) involvement into classical NLS-GFP and alpha-karyopherin independent nuclear transport pathways.
5. Discovery and characterization of a novel leucine-rich export signal in Ssb1p (Hsp70p of yeast).
6. Novel method to study dynamics of the eukaryotic NPC, physiological control of the nuclear dynamics.
7. Discovery and visualization of the NPC exclusion from the Nuclear-Vacuole Junctions and NPCs role in chromatin anchoring.
8. Discovery of the increase in NPC permeability in yeast during the earlier onset of programmed cell death induced by reactive oxygen species (ROS).
9. Discovery of the evolutionary progression of the ribosome decoding center toward more stringent selection of the template for the protein synthesis in the line of prokaryotes-low eukaryotes-higher eukaryotes
TECHNICAL SKILLS
Biology: Xenopus laevis oocytes maintenance and microinjections; obtaining poly- and monoclonal antibody.
Biochemistry: protein expression and purification, enzymes activity, kinetic analysis, DNA, mRNA, tRNA, ribosomes purification and analysis. Developing and improving protocols for purification and analysis of protein complexes, any in vitro biochemistry.
Molecular biology and genetics: in vitro transcription and cell-free translation systems for pro- end eukaryotes; gene cloning, knockout and replacement; two-hybrid system and all standard genetic approaches in yeast and bacteria, including plasmids construction, all types of PCR, site-directed mutagenesis, cloning, sequencing, Western, Northern blots etc.
Cell biology: protein expression and localization in mammalian cell cultures and yeast using indirect immunofluorescence in vitro and living colors in vivo; FRET, Flow cytometry, top expertise in fluorescent and confocal laser microscopy; image and sequence analysis; developing and improving methods for kinetic and physiological studies in vivo.
